چکیده

The metal centres in metalloenzymes and molecular catalysts are responsible for the rearrangement of atoms electrons during complex chemical reactions, they enable selective pathways charge spin transfer, bond breaking/making formation new molecules. Mapping electronic structural changes at sites reactions gives a unique mechanistic insight that has been difficult to obtain date. development X-ray free-electron lasers (XFELs) enables powerful probes structure dynamics advance our understanding metalloenzymes. ultrashort, intense tunable XFEL pulses spectroscopic studies metalloenzymes, under functional conditions real time. In this Technical Review, we describe current state art spectroscopy XFELs highlight some techniques currently development. With more facilities starting operation planning or construction phase, capabilities expected, including high repetition rate, better pulse control advanced instrumentation. For first time, it will be possible make real-time movies solution, while taking place. offer unprecedented capabilities, with their tunable, short pulses. This Review discusses future use probing time conditions.

Methods development for diffraction and spectroscopy studies of metalloenzymes at X-ray free-electron lasers.

X-ray free-electron lasers (XFELs) open up new possibilities for X-ray crystallographic and spectroscopic studies of radiation-sensitive biological samples under close to physiological conditions. To facilitate these new X-ray sources, tailored experimental methods and data-processing protocols have to be developed. The highly radiation-sensitive photosystem II (PSII) protein complex is a prime...

X-ray free-electron lasers

In a free-electron laser (FEL) the lasing medium is a high-energy beam of electrons flying with relativistic speed through a periodic magnetic field. The interaction between the synchrotron radiation that is produced and the electrons in the beam induces a periodic bunching of the electrons, greatly increasing the intensity of radiation produced at a particular wavelength. Depending only on a p...

Near Edge X-ray Absorption Fine Structure Spectroscopy with X-Ray Free-Electron Lasers

We demonstrate the feasibility of Near Edge X-ray Absorption Fine Structure (NEXAFS) spectroscopy on solids by means of femtosecond soft x-ray pulses from a free-electron laser (FEL). Our experiments, carried out at the Free-Electron Laser at Hamburg (FLASH), used a special sample geometry, spectrographic energy dispersion, single shot position-sensitive detection and a data normalization proce...

Toward Single-particle Bioimaging Using X-ray Free-electron Lasers

In this paper we review the recent progress toward single-particle imaging of biological molecules at x-ray free-electron laser (XFEL) facilities. We describe the progression from biological imaging at synchrotrons to imaging at XFELs, discuss recent successes, and point out specific challenges associated with imaging at XFEL facilities.

Structural enzymology using X-ray free electron lasers

Mix-and-inject serial crystallography (MISC) is a technique designed to image enzyme catalyzed reactions in which small protein crystals are mixed with a substrate just prior to being probed by an X-ray pulse. This approach offers several advantages over flow cell studies. It provides (i) room temperature structures at near atomic resolution, (ii) time resolution ranging from microseconds to se...
